Orrit ontdekte dat 1 molecuul voldoende fluorescenrende straling afgeeft om het waar te nemen wanneer het wordt beschenen door laserlicht van een bepaalde golflengte. Orrit ontdekte dat 1 molecuul voldoende fluorescenrende straling afgeeft om het waar te nemen wanneer het wordt beschenen door laserlicht van een bepaalde golflengte. Er wordt als het ware 1 kleur licht op het molecuul af gestuurd, dat vervolgens een andere kleur licht teruggeeft. Daaruit kan het gedrag van enkele moleculen worden afgeleid en worden die moleculen als het ware ’zichtbaar’.
Inmiddels worden 1-molecuulspectroscopie en optische fluorescentie veel gebruikt in de toegepaste wetenschap. Zo kan met de techniek het genoom van een levend wezen worden ontrafeld. Dat kon in de jaren 90 ook al, maar toen kostte het in kaart brengen van de volgorde van DNA van een mens vele miljoenen dollars. 'Nu is hetzelfde te doen voor nog geen 1000 euro', zegt Orrit. '1 heel genoom kan in een middag worden blootgelegd, dankzij single molecule technieken.'
Die technieken zijn bijvoorbeeld toegepast om vast te stellen waar de cholera-uitbraak in Haïti in 2010 vandaan kwam. De ziekte heerst al ruim 5 jaar in het land en heeft duizenden slachtoffers geëist. Beweerd werd dat de epidemie door Nepalese VN-militairen het land binnen was gebracht. Orrit: 'Er zijn toen door het Amerikaanse bedrijf Pacific Biosciences monsters van de bacterie genomen waarna in een weekend het DNA in kaart werd gebracht.'
