Green Light Leiden
Voor chirurgen is het in veel gevallen lastig om tijdens de operatie onderscheid te maken tussen kwaadaardig- en gezond weefsel, met name na behandeling met chemotherapie en/of bestraling voorafgaand aan de operatie en hierdoor neemt de kans toe dat er kwaadaardig weefsel achterblijft.
Onze Green Light Leiden-onderzoeksgroep werkt aan een beeldgeleide techniek om chirurgen te ondersteunen om tijdens operaties dit onderscheid tussen kwaadaardig en gezond weefsel goed te kunnen maken. Dit doen we door met een fluorescente stof en een geavanceerd camerasysteem de tumorcellen tijdens de operatie te laten oplichten. De chirurg kan dan aan de hand van het fluorescentie beeld precies zien waar het kankerweefsel zich bevindt en hierdoor hopen we de vooruitzichten voor de patiënt sterk te verbeteren.
De ontwikkeling van deze tumor specifieke fluorescente tracers vindt plaats in samenwerking met de Faculty of Science (computational drug discovery), afdeling Klinische Farmacie en Toxicologie, de afdeling Cel- en Chemische Biologie en het Centre for Human Drug Research (CHDR). Door deze samenwerking is klinische translatie van het laboratorium naar de kliniek mogelijk. Het doel is om de komende jaren een chirurgische ToolBox te ontwikkelen om zowel kwaadaardige als normale (b.v. zenuw / urineleider) structuren zichtbaar te maken. Tevens hopen we de nieuw ontwikkelde tumor specifieke tracers ook voor therapie en diagnostiek verder te ontwikkelen.
Wetenschappelijke carrière
Na het behalen van mijn doctoraalexamen heb ik promotieonderzoek verricht (verdediging september 1999:“Treatment of Liver Metastases of Colorectal Cancer by Isolated Hepatic Perfusion. Clinical and Experimental Studies on Glutathione Modulation” bij de afdeling Heelkunde en de afdeling Toxicologie van het LACDR naar geïsoleerde lever perfusie als behandeling voor patiënten met uitzaaiingen van dikke darmkanker.
Tijdens mijn opleiding Heelkunde heb ik in 2007 een onderzoek reisbeurs ontvangen van de Nederlandse vereniging van Gastro-intestinale chirurgie om de pionier op het gebied van beeldgeleide chirurgie, John Frangioni, in Harvard (Beth Israel Deaconess Medical Center) te bezoeken. Dit bezoek was het begin van mijn onderzoekslijn en uiteindelijk mijn huidige leerstoel.
