Summary

The research presented in this thesis demonstrated multidisciplinary cooperation among Oncology, Bioengineering, Nanomedicine, and Pharmacology. This work aims to elucidate aspects of Cancer-associated fibroblast (CAF) heterogeneity using 3D human skin equivalents (HSEs) that mimic different types of skin (epithelial) cancers, and to examine the potential of 3D HSEs for drug efficacy evaluation, with a particular focus on tumor-stroma interactions. In this research, we identified specific CAF precursor cells, and explored different CAF-targeted therapeutics by using our 3D HSE system. Our work could provide a stable modeling platform for future tumor-stroma interaction studies, and help to discover new therapeutics for skin cancer patients in the future.
